How to create a bitmap mask?

Hi, I need to work with some bitmap images with a high grade of detail. And I want to use the black information for make a mask where I will project other information.

There I leave an image that can explain better my objectives. I want to leave the white white, and in the dark zones change the color, project a video, etc...
